Sphingomyelin phosphodiesterase (or sphingomyelinase) is a phosphodiesterase enzyme which acts upon sphingomyelin.

It is classified under EC number EC 3.1.4.12.

A deficiency in this enzyme is associated with Niemann-Pick disease.
